The results for polyclonal antibodies were inconsistent. Most polyclonal antibodies did not show sufficient specificity to be used for measuring MLH1 and MSH2 levels. Those that did work did not produce consistent results; thus, we were unable to use them for quantitative detection of these proteins (data not shown). However, we found that two of the monoclonal antibodies (No. 1 and 2 in Table 1) can quantitatively detect full-length MLH1 and MSH2 proteins and which could be combined in a multiplex fashion to detect both proteins in a single assay.
